چکیده
Background: To explore the association of COL2A and Aggrecan polymorphisms and the susceptibility of intervertebral disc degeneration (IVDD). Methods: IVDD patients (n = 354) in Spinal Surgery department of our hospital between June 2010 and June 2013 were allocated to the case group. Healthy people (n = 310) were allocated to the control group. The distribution frequency of COL2A and Aggrecan were detected by using PCR-RFLP. Haplotype analysis and spearman rank correlation analysis were conducted. Results: Statistical significance was found in COL2A (rs2276454) between case group and control group (both P < 0.05), while no statistical significance was found in rs2070739 (both P > 0.05); subgroup analysis on rs2070739 demonstrated the statistical significance between the male patients and controls (both P < 0.05). Statistical significance was found in VNTR between the two groups (all P < 0.05). The frequency of TTL was significantly lower in case group than in control group; the frequency of CTS, CCS was significantly higher in case group than in control group (all P < 0.05). COL2A (rs2276454) was correlated with Schneiderman level, the number of degenerative segments, herniation type and the number of herniated segments (all P < 0.05); rs2070739 was only correlated with Schneiderman level (P < 0.05); Aggrecan polymorphism (VNTR) was associated with Schneiderman level, the number of degenerative segments and the number of herniated segments (all P < 0.05). Conclusion: The COL2A and Aggrecan polymorphisms were associated with the susceptibility of IVDD; TTL, CTS and CCS might be the risk factors of IVDD.

Association of Rs1676486 genetic polymorphism and lumbar disc degeneration in Iranian population
Background: lumbar disc degeneration is a multifactorial degenerative disease which is affected by genetic inheritance and environmental factors. Type XI collagen is important for organization of the extracellular matrix and cartilage collagen construction. Rs1676486 is a SNP that causes the conversion of C-T, resulting in a change in the expression of the collagen 11 alpha chain. The T allele ...
متن کاملVitamin D Receptor Gene, Matrix Metalloproteinase 3 Polymorphisms and the Risk of Intervertebral Disc Degeneration Susceptibility: Meta-Analysis
Several studies have evaluated the association between vitamin D receptor, matrix metalloproteinase 3 (MMP-3) polymorphisms and the risk of intervertebral disc degeneration susceptibility. The findings were inconsistent. This meta-analysis aimed to systematically assess the association between vitamin D receptor, MMP-3 polymorphisms and the risk of intervertebral disc degeneration susceptibilit...
متن کاملOestrogen and parathyroid hormone alleviate lumbar intervertebral disc degeneration in ovariectomized rats and enhance Wnt/β-catenin pathway activity
To investigate the mitigation effect and mechanism of oestrogen and PTH on disc degeneration in rats after ovariectomy, as well as on Wnt/β-catenin pathway activity, thirty 3-month-old rats were ovariectomized and divided into three groups. Ten additional rats were used as controls. Eight weeks later, the rats were administered oestrogen or PTH for 12 weeks, and then discs were collected for te...
متن کاملThe role of proteoglycans in aging, degeneration and repair of the intervertebral disc.
The ability of the nucleus pulposus of the intervertebral disc to resist compressive loads is due to its high content of the proteoglycan aggrecan. Degeneration of the intervertebral disc is preceded and accompanied by a loss of aggrecan due to proteolysis. Biological repair of intervertebral disc degeneration should strive to restore aggrecan content to its optimal functional level. One approa...
متن کاملThe involvement of aggrecan polymorphism in degeneration of human intervertebral disc and articular cartilage.
The functions of the intervertebral disc and of articular cartilage are intimately related to their aggrecan content. Aggrecan is a proteoglycan that interacts with hyaluronan to form large aggregates, which are responsible for the ability of the tissues to resist compressive loads. This function is related to the structure of aggrecan, and in particular to the large number of chondroitin sulph...
